The takeaway
In ATTAIN-2, a daily orforglipron tablet produced more weight loss than placebo over 72 weeks. All participants had type 2 diabetes and received lifestyle support.
What changed?
The trial randomized 1,613 adults with overweight or obesity. Average weight reductions are shown below.
These are the primary estimates, accounting for treatment interruptions and other events. These averages are not individual predictions. Blood-sugar control also improved.
Percent of starting weight; daily study doses

| Group or measure | Result |
|---|---|
| Orforglipron 6 mg | 5.1% |
| Orforglipron 12 mg | 7.0% |
| Orforglipron 36 mg | 9.6% |
| Placebo | 2.5% |
What were the trade-offs?
Digestive symptoms were common. Side effects led 6.1–9.9% of orforglipron participants to stop treatment, versus 4.1% with placebo.
Ten deaths occurred: six across the three orforglipron groups and four with placebo. These counts alone cannot establish whether treatment changed mortality risk.
What does this mean for you?
This study addresses weight loss specifically in people with diabetes. It did not compare the tablet directly with injectable treatments. Eli Lilly funded the trial.
